TSC Therapeutics: Afinitor and Bio 30

A drug called Afinitor/Certican/Everolimus/RAD-001 from Novartis is an immuno-
suppressor and has been used for heart transplantation in the past, to suppress
the graft-rejection. However, like rapamycin, it was recently found to
block the oncogenic kinase TOR, which is abnormally activated in TSC (tuberous
sclerosis) tumors. Thus, in October of 2010, FDA approved this drug to be
used for TSC tumors in particular SEGAs in brain. This year Michal Yalon's
group in Israel reported that this drug (10 mg daily) clearly shrinks the
SEGAs in TSC patients (1). 10 mg pill of this drug costs around US$200 (each
case containing 30 pills=US$5,500-6,500). Unfortunately, however, this drug
causes at least two side effects: immuno-suppression and hypertension.


Combination (Afinitor and Bio 30) Therapy:

In my opinion, this drug (5 mg daily) along with Bio 30, a NZ propolis extract
(1 ml per 10 kg daily), would be the best solution to conduct the TSC therapy
with little (or perhaps "no") side effect. Why? First of all, Bio 30 causes
no side effect, and boosts immune system and reduces blood pressure if patients
suffer from hypertension. In other words Bio 30 would abolish the two major
side effects of Afinitor. Furthermore, in my Bio 30 trials for NF and TSC
patients, Bio 30 clearly stops seizure (epilepsy) associated with TSC and
improves the cognition which is often impaired in TSC patients. Finally,
TOR requires the oncogenic kinase PAK1 for its full activation, which is
blocked selectively by Bio 30. Thus, reduction of daily dose of Afinitor
to a half (taking 5 mg pill, instead of 10 mg pill), along with Bio 30 would
not only reduce the side effect, but also boost its therapeutics effect
synergistically with anti-TSC activity of Bio 30.


Bio 30 alone Therapy:

The following is my "second" opinion. Since afinitor is very expensive, costing
US$200 daily (per 10 mg) , and causes a number of side effects, it would
not be suitable for the life-long therapy of TSC. Thus, I would recommend
TSC patients, in particular "not wealthy" people, to take Bio 30 alone
(the effective "minimum" daily dose: 1 ml per 10 kg). For Bio 30 therapy
costs only around US$1 daily, and causes no side effect, and our recent
human trials for a few TSC children show that Bio 30 alone can stop the
TSC-induced epilepsy and improve the cognition, clearly suggesting that
Bio 30 alone is able to shrink TSC tumors in brain.
